Apparently undeterred by retaliation from a pro-Second Amendment blogger, the editorial staff at Gannett?s ?Journal News? is at it again.
After creating an uproar for publishing the names and addresses of gun owners in New York?s Westchester and Rockland counties, ?Journal News? announced it would release and publish thenames and addresses of permit holders in Putnam County, according to Reuters.
?Further names and addresses will be added as they become available to a map originally published on December 24 in the White Plains, New York-based Journal News,? the report notes.
The map was included in an article entitled ?The gun owner next door: What you don?t know about the weapons in your neighborhood,? that was released shortly after a shooting spree in Newtown, Conn., claimed 26 lives, including 20 children.
?Some 44,000 people are licensed to own pistols in the three counties, the newspaper said. Owners of rifles and shotguns do not need permits,? Reuters adds.
The newspaper?s decision to publish the names and addresses of permit holders has been met with both praise and criticism.

After the newspaper launched its ?gun owners? map, pro-Second Amendment blogger Robert Cox retaliated by releasing a map with the names & addresses of ?Journal News? employees.
For her part, CynDee Royle, VP of news and editor, stands by the newspaper?s decision to publish private citizens? information.
?We knew publication of the database would be controversial, but we felt sharing as much information as we could about gun ownership in our area was important in the aftermath of the Newtown shootings,? she said.
But Al Tompkins, media critic for the Poynter Institute, writes that the newspaper failed to justify its decision to publish permit holders? personal information.
?If journalists could show flaws in the gun permitting system, that would be newsworthy,? he said. ?Or, for example, if gun owners were exempted from permits because of political connections, then journalists could better justify the privacy invasion.?
He continued, adding that he ?feared the dispute might prompt lawmakers to play to privacy fears,? as Reuters notes.
?The net effect of the abuse of public records from all sides may well be a public distaste for opening records, which would be the biggest mistake of all,? he said.

LONDON (AP) ? It is not often that the president of the United States needs to seek fashion advice.
But when Ronald Reagan was getting ready for a visit to England as a guest of Queen Elizabeth II in June 1982, his people had an important question for the Brits: Just what does one wear to go riding with the queen in the magnificent horse country surrounding Windsor Castle?

The fashion inquiry is but one tidbit contained in nearly 500 pages of formerly Confidential documents relating to the Reagan visit being made public Friday by Britain's National Archives. The dossier shows the British government ? led by Prime Minister Margaret Thatcher ? to be extraordinarily interested in pleasing the relatively new president on his two-day visit. British leaders also fretted that perennial cross-Channel rivals might triumph in the tug-of-war for presidential face time in a visit that had to be sandwiched between two summits on the European mainland.
The papers show that top Reagan adviser Michael Deaver had a way of annoying his British counterparts with last-minute changes and requests, and also surprised them with some of his objectives. Deaver, remembered as a shrewd image-builder, said he wanted Reagan to be photographed outside of formal venues, so he wouldn't be seen "exclusively in white tie" at palace functions, even suggesting that Reagan go to a village pub to soak up the atmosphere
There were raised eyebrows, and bruised feelings, when the White House failed to formally reply in a timely fashion to an official invitation from the queen ? the sort of invite that usually commands respect and a prompt reply the world over. The queen's invite was left to languish for weeks and weeks, something that the British believe is simply Not Done.
"It is really for the president to respond to her invitation, which he has not done personally, something that I have pointed out several times here," writes Nicholas Henderson, Britain's ambassador to Washington, in a memo to the British Foreign Office. "As you know those surrounding the president are not deliberately rude: It is simply that they are not well-organized and do not have experience of this sort of thing."
William F. Sittman, a special assistant to Reagan who was involved in planning the trip, told The Associated Press that it is possible the delay in responding to the queen was caused by first lady Nancy Reagan's insistence on consulting her astrologer before travel plans were finalized.
"You have to remember that Mrs. Reagan was very strict about his schedule, and she would consult her astrologer to see if this was the right time to travel," he said. "Sometimes she would back up departures."
The documents make clear that Europe's leaders were desperate for Reagan's attention at a time of high Cold War tensions. A memo from U.K. Cabinet Secretary Robert Armstrong on Feb. 5 expresses concern that a gala, summit-closing dinner at the palace of Versailles outside Paris could delay Reagan's arrival in London. But he warns against pressuring the Reagan entourage to skip the meal at Versailles' Hall of Mirrors because "that would not please the President of the French Republic."
Reagan's aides also worried the British by suggesting the president might have to skip the stop in London because accepting it might anger the Germans, who had offered a similar invitation. But feelings are smoothed over a bit when the Americans assure the British contingent that the Germans are not America's top priority.
"Eagleburger emphasized how much the president himself wanted to go to London," stresses one confidential memo from the British ambassador, referring to senior U.S. diplomat Lawrence Eagleburger. "There should be no doubt about that. Eagleburger also said that at the moment the Germans were not America's favorite allies."
The prospect of a chance to relax from international summitry with a bit of horseback riding with the queen seems to have helped carry the day for the Brits. Asked for the president's favorite type of horse, British planners are told simply that he wants a thoroughbred. He ended up riding Centennial, one of the queen's favorites, and wearing a perfectly fitted sports jacket above his sweater, going for an old-time Hollywood look he carried off with ease.
Much of the actual visit was devoted to pomp and pageantry, or to relaxation, but Reagan did make one speech of consequence. He became the first American president to address a meeting of both houses of Parliament and used the occasion to trumpet his distaste for the Soviet Union, calling it an economic catastrophe.
He said Marxism-Leninism would be left on the ash heap of history ? a prediction that would come to pass in the following decade.

SAN FRANCISCO (AP) - In the private hell of a mother's grief, the sounds come back to Judy Neiman. Her SUV door slamming. The slight bump as she backed up in the bank parking lot. The emergency room doctor's sobs as he said her 9-year-old daughter Sydnee, who previously had survived four open heart surgeries, would not make it this time after being backed over by her Mother's hulking SUV.
Her own cries of: How could I have missed seeing her?
The 53-year-old woman has sentenced herself to go on living in the awful stillness of her West Richland, Wash., home, where she makes a plea for what she wants since she can't have Sydnee back: More steps taken by the government and automakers to help prevent parents from accidentally killing their children, as she did a year ago this month.
"They have to do something, because I've read about it happening to other people. I read about it and I said, 'I would die if it happens to me,'" Neiman says. "Then it did happen to me." There is, in fact, a law in place that calls for new manufacturing requirements to improve the visibility behind passenger vehicles to help prevent such fatal backing crashes, which the government estimates kill some 228 people every year - 110 of them children age 10 and under - and injures another 17,000.
Congress passed the measure with strong bipartisan backing, and Republican President George W. Bush signed it in 2008.
But almost five years later, the standards have yet to be mandated because of delays by the U.S. Department of Transportation, which faced a Feb. 28, 2011, deadline to issue the new guidelines for car manufacturers. Transportation Secretary Ray LaHood has pushed back that deadline three times - promising in February that the rules would be issued by year's end.
With still no action, safety advocates and anguished parents such as Neiman are asking: What's taking so long to remedy a problem recognized by government regulators and automakers for decades now?
"In a way, it's a death sentence, and for no good reason," said former Public Citizen president Joan Claybrook, who once directed the federal agency responsible for developing the rules. The proposed regulations call for expanding the field of view for cars, vans, SUVs and pickup trucks so that drivers can see directly behind their vehicles when in reverse - requiring, in most cases, rearview cameras and video displays as standard equipment.
The National Highway Traffic Safety Administration, charged with completing the new standards, declined requests to discuss the delays. Spokeswoman Karen Aldana said the agency would not comment while the rulemaking process was ongoing but was on track to meet LaHood's latest cutoff date. In a letter to lawmakers in February, LaHood said his agency needed more time for "research and data analysis" to "ensure that the final rule is appropriate and the underlying analysis is robust."
Others insist the issue is money, and reluctance to put any additional financial burdens on an industry crippled by the economic crisis. Development of the new safety standards came even as the Obama administration was pumping billions of dollars into the industry as part of its bailout package.
"They don't want to look at anything that will cost more money for the automobile industry," said Packy Campbell, a former Republican state lawmaker from New Hampshire who lobbied for the law.
"This about money, but it is also about politics," says AOL Autos editor-in-chief David Kiley. "The Obama White House has been lobbied hard by automakers to either delay or eliminate the regulation because of costs the automakers are incurring to meet the new fuel economy regulations."
NHTSA has estimated that making rear cameras standard on every car would add $58 to $88 to the price of vehicles already equipped with dashboard display screens and $159 to $203 for those without them.
The Alliance of Automobile Manufacturers, a lobbying group that represents automakers, puts the total cost to the industry at about $2 billion a year that would largely be passed on to consumers. Last December, the group met with White House budget officials to propose a less expensive alternative: reserving cameras for vehicles with extra-large blind zones and outfitting the rest with curved, wide-angle exterior mirrors.
Before the delays, all new passenger vehicles were to carry cameras and video displays by September 2014. The industry has now asked for two more years after the final rules are published to reach full compliance.
Despite its resistance, the industry on its own has been installing rearview cameras, a feature first popularized two decades ago in Japan and standard on nearly 70 percent of new cars produced there this year. In the United States, 44 percent of 2012 models came with rear cameras standard, and 27 percent had them as options, according to the automotive research firm Edmunds. They come either as standard equipment on premium and luxury vehicles, or as options on lower priced vehicles.
Nine in 10 new cars had console screens available, according to market research firm iSuppli, which would put the price of adding a camera on the low end of the NHTSA's estimates. These backing crashes are hardly a new phenomenon. Emergency room doctors, the National Centers for Disease Control and Prevention and the NHTSA have produced dozens of papers on the problem since the 1980s.
Researchers at the University of California, Irvine, started looking into the issue in the 1990s after noticing toddlers showing up in hospital databases of injured child pedestrians. They found that many of those children had been killed or hurt by vehicles backing out of home driveways. In 1993, the NHTSA sponsored several studies that noted the disproportionate effect of backup accidents on child victims. One report explored sensors and cameras as possible solutions, noting the accidents "involve slow closing speeds and, thus, may be preventable." Still another 1993 report estimated that 100 to 200 pedestrians are killed each year from backing crashes, most of them children.
Three years later, Dee Norton, a reporter at The Seattle Times, petitioned the NHTSA to require improved mirrors on smaller commercial trucks and vans after his 3-year-old grandson was killed by a diaper delivery truck that backed over him.
Adding to the scrutiny were studies by Consumer Reports magazine, which started measuring "blind zones" to determine how far away a toddler-sized traffic cone had to be before a driver looking though the rear window could see it. The research found an overall trend of worsening rear visibility - due in part to designs favoring small windows and high trunk lines, said Tom Mutchler, the magazine's automotive engineer.
"Cameras are basically the only technology that is going to let you see something right behind the bumper," he said.
While no one doubts that cameras could help reduce deaths, they aren't regarded as a perfect solution either. One recent study by a researcher at Oregon State University found that only one in five drivers used a rearview camera when it was available, but 88 percent of those who did avoided striking a child-sized decoy.
In its proposed rule, the NHTSA estimated that rearview video systems could substantially reduce fatal backing crashes - by at least 95 a year - and result in at least 7,000 fewer injuries.
Judy Neiman's 2006 Cadillac Escalade didn't have any cameras installed. They weren't added as an optional package until the following model year. Instead, her vehicle was equipped with a "rear parking assist system" - bumper sensors, an alarm and lights that are supposed to go off within five feet of objects or people. Neither Neiman nor the 10-year-old neighbor boy who had accompanied her and her daughter to the bank on Dec. 8, 2011, would recall hearing any alert, according to a police report.
Sydnee was carrying her purple plastic piggy bank and account book, so she could deposit $5 from her weekly allowance. After the transaction, Neiman slid behind the wheel and waited for the children. She heard the door slam, then saw the boy sitting on the right side of the back seat as she put the car into reverse.
She figured Sydnee was seated behind the driver's seat. Instead, the boy had gotten in first, telling Sydnee to go around and get in from the left side. He would later tell a police investigator that the girl had dropped her piggy bank on her way around the SUV.
Even if she were upright, at 4-feet-3-inches tall, Sydnee would have been practically invisible through the rear window, the bottom edge of which was a few inches taller than she was. As the first anniversary of her daughter's death passed, Neiman hoped that sharing her story might spare other parents from enduring the pain she feels every day.

GDF SUEZ continues to develop its small scale liquefied natural gas (LNG) activities in North West Europe following the award by Fluxys of additional berthing and storage capacity long term rights at the Zeebrugge LNG terminal.

Those capacities will enable GDF SUEZ to reload small LNG carriers, such as LNG feeders and LNG bunkering vessels that will supply LNG as marine fuel for shipping companies.
LNG as a fuel for ships will develop in the future in a context of increasing pressure on maritime regulatoryconstraints on particle emissions in North Europe, as well as on fuel oil prices. No threat to game sales from Connecticut shooting, says analyst
Cowan & Company analyst Doug Creutz is convinced that the Sandy Hook Elementary School shooting incident will have little to no impact on the sales of video games, even the ones depicting violence.
He assured the worried investors that video games will continue to sell well in the market irrespective of a new legislation, which seems highly likely with West Virginia Democrat senator Jay Rockefeller introducing a bill in the Congress for investigation
of effects of violent games on children and their well-being.
Creutz also stated that the consumer backlash in the wake of tragic Connecticut shooting would be minor at most and will therefore not result in significant losses at all.
?We have received many questions from investors about the potential impact of the recent tragedy in Newtown on our covered universe of video game publisher stocks,? Creutz said. ?While this is a difficult issue to address due to the intense emotions surrounding
the incident, we have attempted to present a summary of the relevant facts herein.?
The Cowan & Company analyst noted that the Supreme Court?s ruling last week, which deemed sales of violent games to children is in direct violation of the First Amendment, has laid down a solid ground for any attempts to regulate sales of video game to be
struck down immediately.
Addressing the bill introduced by Rockefeller in Congress, Creutz contended that majority of the adults have already established an opinion about the effect of video game violence and real world violence and these opinions will not change that easily.
The bill introduced in Congress would task the National Academy of Sciences (NAS) to carry out a comprehensive study and investigation of the effects of violence in video games and other programming and their effects on the minds and well-being of children.
Creutz claimed that retailer checks with Amazon has revealed that first-person shooters such as Call of Duty: Black Ops II and Halo 4 are continuing to do really well in terms of market performance, thus making it quite obvious that the consumer behaviour

SANAA, Yemen (AP) ? Gunmen on motorbikes shot dead two Yemeni army officers in the country's capital Tuesday, and clashes between the military and tribal fighters loyal to al-Qaida in a northeastern province killed three soldiers and 11 tribesmen, officials said.
The violence came as al-Qaida's offshoot in Yemen, which the United States considers the most dangerous branch of the terror network, released a new video message calling on Yemeni Muslims to join jihad, or holy war, against America.
The two army officers were gunned down in separate parts of the capital, Sanaa, security officials said. The two were identified as Col. Fadhl Mohammed Jaber, who was shot outside his home, and Col. Saleem al-Gharbani, who was killed near a Sanaa military facility.
Similar attacks have killed several senior Yemeni military and intelligence officials this year. The government has blamed al-Qaida, saying the militants are waging a retaliation campaign over a U.S.-backed military offensive last summer that managed to push militants out of strongholds in several southern towns.
Military officials said clashes between the army and a tribe affiliated with al-Qaida in the northeastern province of Marib killed at least 11 tribesmen and three soldiers over the past 24 hours. The officials said a tribal chief, supported by al-Qaida elements and disenchanted over money allegedly owed to him by the government, has been leading the attacks on the army.
All officials spoke on condition of anonymity because they were not authorized to talk to reporters.
The province of Marib has seen a spate of clashes recently, mostly between the army and some tribesmen who maintain ties with al-Qaida. The tribesmen have also attacked oil pipelines and power stations in the province.
Two months ago, tribesmen blew up main electricity pylons and sabotaged an oil pipeline, protesting a death sentence against one of their members convicted of belonging to al-Qaida and killing security agents.
Some tribal chiefs are also suspected of being allied with former longtime President Ali Abdullah Saleh. Their attacks appeared to be aimed at undermining the new government.
Al-Qaida's Yemen branch, also known as al-Qaida in the Arabian Peninsula, released a video denouncing the United States for what it described as aggressive political and social policies against Islam.
In the video, reported Tuesday by the SITE Intelligence Group, which monitors militant activities, three Yemeni al-Qaida leaders ? including deputy leader Said al-Shihri and military commander Qassim al-Rimi ? urge Yemenis to join the jihad against an alleged American "occupation" of their country.
The video was produced by al-Qaida's local media arm, the al-Malahem Foundation.

Dec. 23, 2012 ? In a collaborative study published online today in Nature Biotechnology, researchers from Kunming Institute of Zoology, Chinese Academy of Sciences, BGI, and other institutes, have completed the first genome sequence of domestic goat by a robust approach integrated with next-generation sequencing (NGS) and whole-genome mapping (WGM) technologies. The goat genome is the first reference genome for small ruminant animals and may help to advance the understanding of distinct ruminants' genomic features from non-ruminant species. This work also yields a valuable experience for facilitating the de novo assemblies of large, complex genomes in the future.
Goats are recognized as an important member of the world livestock industry, and with many unique biological features. They are an important economic resource in many developing countries around the world, especially in China and India. However, despite their agricultural and biological importance, breeding and genetic studies of goats have been hampered by the lack of a high quality reference genome sequence. The goat genome sequence will be useful for facilitating the identification of SNP markers for marker-assisted breeding, and improving the utility of the goat as a biomedical model and bioreactor.
With the availability of next-generation sequencing (NGS), draft assemblies are easy to generate nowadays. However, to finish a sequence to the chromosome level remains a hard nut to crack. In this study, the results show that a single NGS platform, when combined with whole-genome mapping technology, could produce a finished assembly much faster and with high quality than other currently available mapping strategies such as BACs or FISH. Through this integrated approach, researchers obtained the ~2.66 Gb goat reference genome from a female Yunnan black goat.
Transposable elements (TEs) are major components of mammalian genomes and contribute to gene and/or genome evolution. The TEs in goat genome are similar to those of cattle, and contain large numbers of ruminant-specific repeats, such as SINE-tRNA and SINE-BovA. It is reported that SINE-BovA repeat expanded primarily in the cattle genome. However, in this study, researchers found the SINE-tRNA repeat expanded specifically in the goat genome.
Through constructing a phylogenetic tree among goats, cattle, horses, dogs, opossums and humans, researchers found the goat shared a common ancestor with cattle about 23 million years ago. Further comparison analysis revealed 44 rapidly evolving genes under positive selection, seven of which are immune-related genes and three are pituitary hormone or related genes. The immune-related genes identified also exist in cattle. The findings suggest that the rapid evolution of pituitary hormones may be related to the different features between goat and cattle in milk production, development rates of the fetus and/or hair variation.
The major histocompatibility complex (MHC) plays an important role in the immune system. In this study, the goat MHC was found to be located on chromosome 23 and contains two regions with length of 2.25 Mb and 360 kb, respectively. With the high quality genome assembly, further understanding of the goat MHC will be useful for immunological studies and vaccine development.
One of the distinguishing characteristics of mammals is the protective growth known as hair. It is produced by hair follicles within the skin, which could provide either protection (guard hairs) or insulation (underfur). The two major hair follicles include the primary hair follicle that produces only coat hair in all mammals, and the secondary hair follicle that can produce the cashmere or "fine hair" in certain mammals, including goats and antelopes. Despite a 2,500-year history and the extent of raw cashmere production, people are lack of understanding of the molecular mechanisms of cashmere formation and development.
Researchers conducted transcriptomic analysis on the primary and secondary follicles of a cashmere goat, revealing 51 genes that are differentially expressed between the two types of hair follicles. Keratin and keratin-associated proteins are the main structural proteins of hair fibers, determining the quality of fiber together. In the study, 29 keratin genes and 30 keratin-associated protein genes were detected in both types of follicles. Interestingly, they found two keratin genes and ten keratin-associated protein genes were consistently differentially expressed between primary and secondary hair follicles, suggesting that the keratin-associated protein genes may be more important in determining the structure of cashmere fibers. In addition to the keratin genes and keratin-associated protein genes, researchers also found several enzymes of amino acid biosynthesis, with implications in regulating primary hair growth and hair cycle.
Xun Xu, Deputy Director of BGI, said, "The goat reference genome is an important stepping stone in the molecular breeding of cashmere goats, and will help to advance the comparative studies on ruminants. The transcriptomic analysis on the primary and secondary follicles will open a new way for better improving the quality cashmere wool."

CAMBRIDGE, Mass. (December 23, 2012) In the perilous hours immediately after birth, a newborn mammal must survive the sudden loss of food supply from its mother. Under normal circumstances, newborns mount a metabolic response to ward off starvation until feeding occurs. This survival response involves a process of controlled breakdown of internal energetic sources known as autophagy. Although autophagy has been well documented, the key mechanistic regulators of autophagy in vivo have remained poorly understood.
Whitehead Institute researchers have discovered that a family of nutrient-sensing enzymes, dubbed Rag GTPases, modulates the activity of the mTORC1 protein complex, whose inhibition is essential for autophagy and survival in newborns. The finding, reported this week in the journal Nature, emerges from the lab of Whitehead Member David Sabatini, whose earlier in vitro studies showed that mTORC1 (for "mechanistic target of rapamycin complex 1") senses the presence of vital amino acids via interactions with Rag GTPases.
To assess the impact of this Rag GTPase-mTORC1 relationship in mammals, the lab generated mice genetically altered to continually express an active form of the GTPase RagA and compared them with wild-type mice. In normal mice, RagA is activated in the presence of nutrients, and turns on the mTORC1 pathway, which regulates organismal growth in response to nutrient availability. If the mice are deprived of nutrients, RagA is switched off, deactivating mTORC1 and initiating autophagy to tide the animal over until the next feeding. However, in the altered mice, RagA's continuous activity keeps mTORC1 active, despite a dearth of available nutrients. Instead of mTORC1 triggering autophagy, the animals' metabolisms remain unchanged, resulting in nutritional crisis and death.
"What happens to a newborn animal with the RagA enzyme always on is pretty shocking," says Sabatini, who is also a professor of biology at MIT and a Howard Hughes Medical Institute (HHMI) investigator. "A normal neonate animal within an hour after birth responds to that condition, but one with its RagA stuck 'on' doesn't, and it dies. It basically has a huge energetic and nutritional crisis because it can't make the adaption."
These striking results stunned Alejo Efeyan, a postdoctoral researcher in the Sabatini lab, and first author of the Nature that describes this work.
"We were surprised that there was no inhibition of this pathway independent of RagAthat there is no backup system," says Efeyan. "And that RagA is a more global nutrient sensor that goes beyond its known function as an amino acid sensor."
RagA's role as an amino acid sensor had been established in cultured cells by the Sabatini lab. Yet when Efeyan compared nutrient levels in fasting newborn RagA-active mice with those of fasting pups with normal RagA, not only amino acids were reduced in RagA-active animals, also glucose levels were dangerously low. The animals were unable to "sense" either of these reductions, so autophagy failed to initiate in the RagA-active pups, which all died within hours of birth.
This newly identified function for RagA suggests much remains unknown about the cell biology of nutrient sensing, an area of research that Sabatini and his lab continue to investigate.

CAMBRIDGE, Mass. (December 23, 2012) In the perilous hours immediately after birth, a newborn mammal must survive the sudden loss of food supply from its mother. Under normal circumstances, newborns mount a metabolic response to ward off starvation until feeding occurs. This survival response involves a process of controlled breakdown of internal energetic sources known as autophagy. Although autophagy has been well documented, the key mechanistic regulators of autophagy in vivo have remained poorly understood.
Whitehead Institute researchers have discovered that a family of nutrient-sensing enzymes, dubbed Rag GTPases, modulates the activity of the mTORC1 protein complex, whose inhibition is essential for autophagy and survival in newborns. The finding, reported this week in the journal Nature, emerges from the lab of Whitehead Member David Sabatini, whose earlier in vitro studies showed that mTORC1 (for "mechanistic target of rapamycin complex 1") senses the presence of vital amino acids via interactions with Rag GTPases.
To assess the impact of this Rag GTPase-mTORC1 relationship in mammals, the lab generated mice genetically altered to continually express an active form of the GTPase RagA and compared them with wild-type mice. In normal mice, RagA is activated in the presence of nutrients, and turns on the mTORC1 pathway, which regulates organismal growth in response to nutrient availability. If the mice are deprived of nutrients, RagA is switched off, deactivating mTORC1 and initiating autophagy to tide the animal over until the next feeding. However, in the altered mice, RagA's continuous activity keeps mTORC1 active, despite a dearth of available nutrients. Instead of mTORC1 triggering autophagy, the animals' metabolisms remain unchanged, resulting in nutritional crisis and death.
"What happens to a newborn animal with the RagA enzyme always on is pretty shocking," says Sabatini, who is also a professor of biology at MIT and a Howard Hughes Medical Institute (HHMI) investigator. "A normal neonate animal within an hour after birth responds to that condition, but one with its RagA stuck 'on' doesn't, and it dies. It basically has a huge energetic and nutritional crisis because it can't make the adaption."
These striking results stunned Alejo Efeyan, a postdoctoral researcher in the Sabatini lab, and first author of the Nature that describes this work.
"We were surprised that there was no inhibition of this pathway independent of RagAthat there is no backup system," says Efeyan. "And that RagA is a more global nutrient sensor that goes beyond its known function as an amino acid sensor."
RagA's role as an amino acid sensor had been established in cultured cells by the Sabatini lab. Yet when Efeyan compared nutrient levels in fasting newborn RagA-active mice with those of fasting pups with normal RagA, not only amino acids were reduced in RagA-active animals, also glucose levels were dangerously low. The animals were unable to "sense" either of these reductions, so autophagy failed to initiate in the RagA-active pups, which all died within hours of birth.
This newly identified function for RagA suggests much remains unknown about the cell biology of nutrient sensing, an area of research that Sabatini and his lab continue to investigate.
